Every computer with an industry-standard sound card is able to record from a variety of sources: a microphone, stereo, CD player, line-in, etc. To record an audio clip playing on you computer, you will need to record from stereo. Here's how:

Go to Start > Control Panel > Sounds and Audio Devices
Click on the Audio tab
Click on the Volume button in the Sound Recording group
Go to Options > Properties
Check the box next to 'Stereo Mix' under 'Show the following volume controls:' and click OK.
Check 'Select' next to Stereo Mix
Drag the Stereo Mix volume control all the way up.

Now you need to a program to record the audio. The Sound Recorder program that comes with Windows works fine, but feel free to use other programs like Adobe Audition. You are now ready to begin recording. Click the record button in Sound Recorder and play the portion of clip you wish to capture. Good luck.
